Abstract
人口老齡化是現今社會不可擋的趨勢,如何延緩改善生理機能的退化
及老化是全球重要的課題。軟骨素已被發現具有許多生物活性,例如抗凝
血、抗發炎和抗氧化等。目前尚未有軟骨素於老化方面的相關研究,然而軟骨素的抗氧化能力顯示其可能具有延緩老化的潛力,因此本論文旨在探討鱘龍魚軟骨萃取物對於老化促進小鼠 (senescence accelerated mouse prone-8,SAMP8) 之老化表徵和功能評估與相關機制探討。
首先應用運用鱘龍魚頭骨與中骨進行萃取,所得之萃取物與SAMP8
小鼠骨髓和脂肪間葉幹細胞作為體外老化細胞測試平台。結果顯示鱘龍魚軟骨萃取物可增加抗氧化酵素Superoxide dismutase, Catalase 和Glutathioneperoxidase 的活性,以及抑制老化相關的脂肪分化,並增加硬骨與軟骨分化能力,也抑制了老化染色β-半乳糖苷酶的表現。而動物實驗方面,活體測試進行了生物活性測試與生存試驗。生物活性測試以6 月齡雌性SAMP8 小鼠為對象,分為控制組與實驗兩組,每組10 隻,共2 組,以0.492 mg/g BW 鱘龍魚軟骨萃取物餵食4 個月後進行分析,結果顯示,鱘龍魚軟骨萃取物可提升SAMP8 小鼠體內血液、腦、心臟、肝臟的抗氧化酵素Superoxidedismutase, Catalase 和Glutathione peroxidase,並透過Micro-CT 發現鱘龍魚軟骨萃取物可減緩SAMP8 小鼠的骨質流失。此外,基因檢測發現鱘龍魚軟骨萃取物可改善小鼠幹細胞的老化基因表現,包括降低老化基因P16、P19、P53 的表現,並提高Sirt1 與c-fos 等延緩老化基因表現。生存試驗則以10 月齡雌性SAMP8 小鼠,同樣分為控制與實驗兩組,每組10 隻,以管餵0.492mg/g BW 鱘龍魚軟骨萃取物進行存活率試驗,結果顯示,鱘龍魚軟骨萃取物
可顯著的延長小鼠的月齡 (對照組/實驗組:11.72352±0.66702 /
15.53±2.36437)
本研究證實鱘龍魚軟骨萃取物可於體外促進幹細胞的活性及能力並減
緩其老化表現,進一步發現可增進小鼠體內器官的抗氧化能力、改善相關老化表徵並延長小鼠壽命,綜合以上結果顯示鱘龍魚軟骨萃取物具有延緩老化的能力。
