前瞻研究開創先機
臺北醫學大學醫科所博士班李嘉華同學在附屬雙和醫院吳志雄院長及北醫醫技系何元順教授的指導下,研究成果登上國際期刊Journal of the National Cancer Institute (JNCI;IF:14.069)。在吳院長暨何教授領導的癌症研究團隊證實,香菸中尼古丁可藉由特定之尼古丁受體(a9-nAChR)作用,與國人乳癌形成有密切關聯。這項研究利用real-time PCR分析276例國人乳癌組織(圖A),證實尼古丁受體的表現較正常組織明顯高約為 8倍(圖B),證實尼古丁受體的高表現量與國人乳癌致癌作用有相關性。
在活體實驗中,研究團隊也證實將乳癌細胞(MDAMB-231)之尼古丁受體抑制(knock-down)其表現,將會影響細胞株於老鼠的腫瘤生長速度。反之,將尼古丁受體過度表現之MCF-10A-Nic(DOX) 細胞轉殖入免疫缺陷鼠(nude mice)內。尼古丁受體過度表現(DOX-老鼠在腫瘤生長速度上明顯較DOX+老鼠快許多。)
未來癌究團隊將希望探討乳房細胞「早期癌變」之基因表現。因此、除了探討a9-nAChR基因在這些「早期癌變」細胞之表現,也將探討在「早期癌變」細胞中,乳癌幹(stem)細胞之表現情形。探討香菸所誘發之乳癌致癌機制,針對國人乳癌致癌因子提供一比較明確的研究方向。
