Abstract
長期高熱量高脂肪的攝取易引致肥胖、血脂異常等問題,並造成心血管疾病的發生。血管病變或嚴重肢體缺血(critical limb ischemia, CLI)為肥胖的併發症。內皮前驅細胞(endothelial progenitor cell, EPC)為骨髓來源的細胞,具有幹細胞特性能夠分化成內皮細胞,血液中的EPC可以修復受損的內皮細胞,使血管內皮細胞能夠維持其功能及完整性。麩醯胺(Glutamine, GLN)為快速生長細胞之能量來源,在代謝壓力下被認為是條件性的必需胺基酸。本實驗目的為探討飲食中給予GLN對肥胖小鼠下肢缺血後,血液中EPC調動及脂肪組織adipokines分泌之影響。本實驗共分為四大組,分別為Normal control (NC)組、High fat control diet (HC)組、High fat diet + Ischemia (HI)組、High fat diet + GLN + Ischemia (HG)組,又以犧牲時間點分為1天及7天犧牲組。實驗期間NC組給予標準囓齒類動物飼料、HC、HI組採用熱量60%由脂肪提供之高脂飼料、HG組給予GLN含量占總氮量之25%的高脂飼料。本篇研究結果顯示給予GLN組,下肢缺血一天後血中EPC比例增加,七天後增加肌肉組織hypoxia inducible factor-1α、matrix metalloproteinase-9 (MMP-9) mRNA表現及血漿中MMP-9濃度,亦能夠減少脂肪組織tumor necrosis factor-α、interleukin-1β、monocyte chemoattractant protein-1表現,推測GLN添加對於下肢缺血之肥胖小鼠將有助於EPC調動至缺血組織並減少脂肪組織之發炎反應。
