Abstract
􏰀乳癌可藉由其細胞來源及其細胞膜受體之表現量而作分類。其中,三陰性乳癌缺乏雌激素受體(ER)、孕激素受體(PR)以及人類表皮生長因子第二型受體(HER2)的表現,使其在治療選擇上受限,因而導致三陰性乳癌病人不好的預後。三陰性乳癌中有著類胰島素生長因子受體(IGF-1R)的高表現量。類胰島素生長因子受體被實驗證明與癌細胞之上皮間質轉化、細胞移行、組織侵犯及癌幹細胞特性有相關。雖然類胰島素生長因子受體在乳癌病人研究中有大量記載,然而其中對於三陰性乳癌細胞的調控機轉尚未明瞭。醣化作用在蛋白質後轉譯修飾中扮演非常重要的角色,其功能在於修飾與調控受體下游訊息傳遞路徑。我們推測類胰島素生長因子受體的醣化作用可能藉由影響不同的細胞訊息傳遞路徑來達到三陰性乳癌細胞移行的能力。因此,本論文除了探討類胰島素生長因子受體對於三陰性乳癌細胞移行的角色,更進一步嘗試釐清類胰島素生長因子受體藉由後轉譯醣基化修飾調控三陰性乳癌細胞移行的角色。藉由釐清類胰島素生長因子受體之後轉譯醣基化修飾及其下游傳遞路徑從而發展出臨床上對於三陰性乳癌嶄新的治療方式。
