Abstract
  痤瘡是一種常見的皮膚疾病,受性荷爾蒙的影響,好發在青春期,其形成的原因包括:雄性荷爾蒙及皮脂分泌旺盛、皮膚毛孔角化異常、毛囊內細菌過度增殖及發炎反應。近年來,從海洋生物中分離出了10000多種海洋天然化合物,其中海綿及珊瑚都是最具生產力的海洋生物,由於目前文獻中尚未發現能有效的治療痤瘡,因此本研究將從海綿及珊瑚中,探索具抗痤瘡潛力之起始天然物。
  利用紙錠瓊脂擴散試驗之抗菌活性分析、脂多醣(Lipopolysaccharide;LPS, 500 ng/ml)及熱處理痤瘡桿菌(300 μg/ml)誘導 RAW264.7 巨噬細胞及 HaCaT 角質細胞之發炎反應,評估13種海洋天然物抗菌功效及抗發炎作用機轉,並利用DHT(1000 nM)誘導HaCaT 角質細胞之皮脂分泌,評估抑制脂質合成途徑之機轉。結果顯示:13種天然物中11-dehydrosinulariolide(SC-2)、3,4:8,11-Bisepoxy-7-acetoxycembra-15(17)-en-1,12-olide(SC-7) 及sinularin(SC-9)明顯抑制 NO 的產生,其 IC50 分別為 5.66 ± 0.19, 15.25 ± 0.25和3.85 ± 0.2 μM,也對 iNOS 的表現有顯著的抑制作用,其中又以 SC-9 對 PGE2的產生抑制作用最強,且這三種化合物也明顯抑制 LPS 和熱處理痤瘡桿菌誘導 RAW264.7 和 HaCaT 細胞之 MAPKs 途徑(p-ERK與p-JNK)的表現,具顯著抗炎作用,且皆能抑制 DHT(1000 nM)誘導 HaCaT 細胞 24 小時後產生之皮脂的分泌及脂質合成途徑中p-AKT及SREBP-1的表現量。
  綜合而論,軟珊瑚天然物是值得開發成高經濟價值的藥用天然物,且有未來發展成抗痤瘡外用產品之潛力。
