Abstract
卵巢癌是一種常見的婦科惡性癌症,卵巢癌的發生率在全球女性中排名第八名,死亡率排名第五名,在台灣發生率與死亡人數也呈現逐年增加的趨勢,開發新的治療藥物與策略勢在必行。天然產物在發展疾病治療上有悠久的歷史,而海洋環境的生物多樣性提供了許多新機制應用於疾病的治療。從海綿中分離出的天然海洋化合物 Manzamine A (Manz A) 具有廣泛且良好的生物活性,如抗癌、抗瘧疾、抗發炎等功能。近年來,質譜技術的迅速發展促進了蛋白質體學分析的研究。因此,我們期望透過蛋白質體學的分析方法全面性探討Manz A抑制人類卵巢癌細胞 (OVCAR8和A2780) 的潛在分子機制。在這項研究中,我們驗證Manz A顯著抑制人類卵巢癌細胞株之增殖,同時發現Manz A誘導卵巢癌細胞的細胞週期停滯在G0/G期和細胞凋亡。透過蛋白質體學分析方法,我們從介入Manz A的A2780細胞組別中鑑定到29,222條胜肽,對應到4,073個可量化的蛋白質,其中有119個表現量顯著上升和72個顯著下降的蛋白質。在介入Manz A的OVCAR8細胞組別中鑑定到31,458條胜肽,對應到4,176個可量化的蛋白質,其中有147個表現量上升與96個表現量下降的蛋白質。交集分析在兩株細胞中共有38個表現量上調和19個表現量下調之蛋白質,針對共同表現差異之蛋白質進行功能性富集分析,結果揭示Manz A增加依賴性鐵死亡與細胞自噬作用途徑相關蛋白的表達,並減少了粒線體氧化磷酸化相關蛋白的表達。通過追蹤卵巢癌細胞株內的鐵離子發現Manz A 促進TFRC的表達使細胞內的鐵離子含量增加,而過多的鐵離子部分被轉移到粒線體內。透過粒線體螢光染色觀察到Manz A導致卵巢癌細胞株內粒線體型態腫脹和不完整,且透過脂質過氧化探針證實Manz A增加了卵巢癌細胞株內脂質過氧化程度,並且促進麩醯胺酸轉運蛋白SLC38A1的表現以促使更多的GSH作為內清除氧化壓力的機制,然而GSH生成的速度無法滿足消耗的速度,最終引發依賴性鐵死亡 (Ferroptosis)。總結本研究透過蛋白質體學分析與實驗驗證Manz A透過誘發依賴性鐵死亡抑制卵巢癌細胞的存活,顯示Manz A具有治療卵巢癌的潛力,提供了未來發展治療卵巢癌的藥物的重要基礎。
