Abstract
背景:金黃色葡萄球菌(Staphylococcus aureus)為全球重要臨床致病原菌,現行檢測方法仍仰賴細菌培養與藥敏試驗(AST)。雖具高準確度,但需 2~5 天方能獲得結果,可能延誤治療並加速抗生素抗藥性(AMR)問題。因此,發展快速、低成本且可攜式的即時檢測(POCT)平台,有助於提升臨床感染監測效率與決策精準度。
研究目的:本研究旨在開發一種結合顯色反應與影像分析的雙模式紙基微流體(μPAD)平台,用於金黃色葡萄球菌的快速檢測與半定量分析。透過標準化影像擷取與數位影像處理,量化顏色強度並建立與菌濃度間的指數回歸模型。
材料與方法:μPAD 以人工壓痕方式製備微流道結構,樣本經十倍與對半稀釋(n = 5)後進行抗原–抗體反應與 HRP–TMB 顯色,反應時間固定為 10 分鐘。以標準化光源與距離拍攝影像,並使用 Python/OpenCV 進行灰階轉換、二值化、圓區域定位與 RGB/HSV 色彩量化;其中 RGB 的 B 通道具最高靈敏度。因傳統培養法無法提供連續色彩與濃度資料,故以混淆矩陣進行方法一致性分析。
結果與結論:平台之顯色訊號與 CFU 呈高度相關(R² = 0.96),偵測極限約為 10⁶ CFU/mL。混淆矩陣分析顯示相較於傳統培養法,本平台具有 80% 的整體準確率與 86% 的特異性,且不同濃度條件下皆呈現穩定可重現的顏色反應。
