Abstract
中孔洞二氧化矽奈米 (Mesoporous Silica Nanoparticles, MSN) 顆粒是一種生物相容性材料,目前被廣泛研究用於運送藥物、蛋白質、酵素送入至生物體。然而此材料仍然有一些在用於臨床前需要克服的問題,如在生物體中MSN被快速清除及穿透血腦屏障等等。而我們的研究則是嘗試合成出不同表面電荷的MSN,並利用斑馬魚動物模式嘗試找出有可能穿透血腦障壁的MSN粒子及其特性。我們已合成了不同種類直徑50 nm的MSN,分別具有-40,-20,+ 20,+ 35 mV的表面電荷。MSN透過加入的聚乙二醇 (PEG)以增加親水性,且避免在培養液中聚集。之後,奈米顆粒的微注射到胚胎受精後3 天斑馬魚中,發現該強負電荷的粒子可以廣泛分佈和滲透屏障進入魚腦中。而這些奈米顆粒在細胞毒性測試中,奈米顆粒200 µg/µL的濃度範圍內對兩種腦細胞株U87-MG與CTX-NA2並沒有表現出毒性在。在其中針對有滲透效果的N4-MSN@PEG/THPMP50進行調整THPMP結合多寡或改變粒徑大小利用穿透血腦障壁的差異評估其效果。在藥物裝載研究證明了N4-MSN@ PEG/THPMP50是可以利用中孔洞空間來裝載藥物,在斑馬魚模式中確認N4-R-MSN@PEG/THPMP50滲透到大腦,並於細胞毒性分析中表現出低細胞毒性腦細胞的好材料。
