Abstract
甲狀腺未分化癌 (ATC) 是最致命的惡性腫瘤之一,中位存活期為 3-5 個月,幾乎絕對的疾病特異性死亡率,主要影響老年女性患者。儘管腫瘤微環境具有迅速和致命的性質,但在理解腫瘤微環境方面取得了實質性進展,為創新的全身治療鋪平了道路,旨在提高總生存率,特別是與多模式治療相結合時。儘管如此,這些治療的療效可能受到其相關發病率的限制,這突顯了迫切需要進行更多研究,以在將潛在毒性降至最低的同時獲得更好的結果。USP20 是癌症中一種關鍵的去泛素化酶,可能通過穩定抗腫瘤蛋白或致瘤蛋白來潛在地調節腫瘤發生。最近的突破揭示了它在細胞過程中的作用,支援了以 USP20為主進行標靶治療可能是腫瘤治療的關鍵的觀點。我們選擇 SW1736 和 8305C ATC 細胞系作為細胞模型,用於檢查新型 USP20 抑製劑GSK2643943A的潛在抗癌作用。首先,我們研究了細胞存活率。用不同濃度(0–8μM)的GSK2643943A處理兩個ATC細胞系24小時至72小時。MTT檢測結果表明,GSK2643943A濃度依賴性地抑制了這些 ATC 細胞的細胞存活率。其次,我們研究了細胞黏附。用
GSK2643943A(0–8 μM) 預處理ATC細胞24 小時後,進行黏附測定。細胞附著分析結果表明,GSK2643943A可能略微引起ATC細胞數量的減少。為了進一步研究 GSK2643943A 對細胞運動的影響,將進行細胞凋亡、去黏附和遷移試驗。透過細胞遷移的實驗得知在加入 GSK2643943A後會使 ATC細胞的爬行能力下降,接著透過去黏附的實驗可以發現加入 GSK2643943A會使 ATC細胞的更快的脫落。最後免疫墨點法分析結果顯示,加入GSK2643943A會導致細胞凋亡酵素PARP裂解;促進細胞週期G1/S phase進行之調控相關的主要蛋白E2F1表達量下降。綜合以上所有實驗結果,GSK2643943A可能會藉由誘導ATC細胞的細胞凋亡、降低促進細胞週期 G1/S phase 進行之調控相關主要蛋白來達到抑制細胞存活的作用;透過降低ATC細胞去黏附的能力來減緩細胞遷移的能力。
