Abstract
一、中文摘要 (Chinese Abstract)
過去本實驗室的研究發現,在內皮細胞膜上葉酸的受體與女性荷爾蒙受體會形成複合體,相互影響葉酸及女性荷爾蒙對於血管內皮細胞增生及遷移的作用。當單獨給予葉酸或助孕酮時,都會抑制培養中的血管內皮細胞之增生及遷移現象。然而同時給予葉酸及助孕酮時,則這些抑制現象則完全消失。因為這些發現,讓我們聯想到是否可以利用葉酸來改善女性賀爾蒙在臨床治療上可能產生的副作用。先前的文獻報導指出,助孕酮會與乳癌T47D細胞上的助孕酮受體結合,造成T47D細胞轉移的加速,而使病情惡化。因此首先我們利用RT-PCR的方法來確定T47D細胞上是否同時具有助孕酮及葉酸的受體。接著利用Transwell invasion assay,我們發現單獨給予葉酸或助孕酮皆促進這些乳癌細胞的入侵能力,然而同時給予葉酸及助孕酮時,則這些促進現象則完全消失了。而在Transwell invasion assay的條件下,葉酸及助孕酮的處理都不會影響到乳癌細胞的存活率、對細胞外基質的貼附能力、細胞的增生、以及MMP2及MMP9的活性,顯示葉酸與助孕酮在乳癌細胞上的交互作用是透過對遷移能力的影響。我們也繼續探討葉酸與助孕酮對於乳癌細胞的遷移作用彼此之間影響的作用分子機轉。本研究的發現將有助於未來利用葉酸來改善女性賀爾蒙在臨床治療上可能產生的副作用,特別是乳癌的惡化現象。
