Abstract
哺乳動物中,鳥苷酸環化酶(guanylate cyclase, GC)主要可分為兩類型,分別為水溶性蛋白型鳥苷酸環化酶(soluble guanylate cyclase, sGC)與膜蛋白型鳥苷酸環化酶(particulate guanylate cyclase, pGC),兩者皆具有可將GTP轉換成cGMP的酵素活性。本實驗室在先前的研究中已經證實GC-G(mGC-G)表現於雄性小鼠體內的睪丸組織並且參與在成熟精子的獲能反應中;為了進一步研究GC-G在雌性小鼠生殖系統中的表現,我們先使用陰道抹片的檢查與血清中動情素的濃度來偵測雌性小鼠的動情週期,進一步利用RT-PCR與免疫組織染色法證實mGC-G的mRNA與蛋白主要表現在卵巢中的卵膜及顆粒細胞上,還有各個動情週期之輸卵管及子宮的內膜上;利用卵巢切除的雌鼠動物模式,我們證實mGC-G在小鼠子宮中的表現受到動情素的調控,顯示mGC-G可能為動情素下游的分子。另一方面我們利用初代培養顆粒細胞的細胞模式藉以釐清mGC-G在卵巢中所扮演的角色。利用RT-PCR與細胞免疫螢光染色的實驗結果證實mGC-G在顆粒細胞的表現;進一步利用ECD-Homophilic interaction之細胞作用模式,我們證實mGC-G-ECD-Fc recombinant protein可以活化顆粒細胞的mGC-G而提高細胞內cGMP濃度,並且降低在顆粒細胞中影響合成動情素之Aromatase酵素基因的表現。綜合以上結果,我們可知mGC-G不只表現在雄性小鼠睪丸中,同時也有表現在雌性小鼠的生殖系統中;特別可能在顆粒細胞中扮演促進雌激素生成上的重要角色。
