Abstract
骨骼肌的健康仰賴於骨骼肌幹細胞及肌肉纖維周邊微環境的交互作用。骨骼肌幹細胞的狀態,包含了活化、自我更新、增生、分化,都受到幹細胞內在特性與微環境中外在訊號密切調控,然而微環境調控肌肉幹細胞的詳細機制,目前仍尚未被透徹地探討。其他全面性的變化,像是老化也會影響骨骼肌幹細胞的再生能力,老化造成肌肉質量上及功能上的衰退以及膠原蛋白代償性地堆積在肌肉中,因此關於老化對骨骼肌的調控機制也需要進一步的研究。
核醣核苷酸還原酶M2B(Rrm2b)在提供DNA修復及維持粒線體恆定的原料扮演著重要的角色,過去發現RRM2B的變異和粒線體DNA缺乏症候群及肌肉病變高度相關,並且在Rrm2b全身性基因剔除的小鼠中曾發現骨骼肌萎縮的現象,因此我們假設Rrm2b基因在骨骼肌修復及再生過程中具有關鍵的作用。
本篇論文中,我試圖探討老化及基因缺失對調節骨骼肌再生能力的影響。我們發現老化限制了肌肉中膠原蛋白的分解能力,使得膠原蛋白隨著老化的過程在肌肉中斷地累積。然而在氯化鋇造成肌肉損傷後,老年小鼠可以有效地修復肌纖維並減少肌肉中膠原蛋白的沈積。我們也發現Rrm2b基因在骨骼肌纖維的缺失會影響肌肉幹細胞的狀態。Rrm2b在肌纖維而非幹細胞的特異性缺失導致骨骼肌質量及力量的衰減,同時也引發隨著年齡增加比例的再生肌肉纖維(centrally nucleated myofibers),並造成肌纖維萎縮以及肌肉幹細胞數量減少,從而限制了骨骼肌整體的再生能力。Rrm2b缺失的肌肉纖維同時會分泌一些肌肉激素 (myokines) ,如纖維細胞生長因子21 (Fgf21) 及生長/分化因子15 (GDF15) ,可能促使肌肉幹細胞自發性的分化並消耗整體肌肉幹細胞的數量。此外,這個小鼠模型也表現了某些粒線體肌病(mitochondrial myopathy)的特徵,有機會成為研究相關疾病的動物模型。
這些研究成果提供了部分能解釋老年肌肉纖維化機制的線索,也有助於開發抗纖維化療法來減少老年肌肉中的膠原蛋白沈積。而Rrm2b在肌纖維中的作用,對於調節肌肉幹細胞狀態具有關鍵的功能,這些成果可能有助於建立肌肉疾病的相關治療策略。
