Abstract
發炎性腸道疾病屬於慢性的疾病,包含潰瘍性結腸炎與克隆氏症兩種,當疾病生成時會觀察到發炎、氧化壓力及凋亡反應。目前主要治療的方式為藥物,但長期使用會有副作用生成,而過去研究指出枸杞多醣 (Lycium barbarum polysaccharides, LBP) 與辣椒素 (capsaicin, CAP) 具有影響發炎、氧化及凋亡的效果。因此本次研究欲探討LBP與CAP對於脂多醣 (lipopolysaccharide, LPS) 誘導人類結腸腺癌細胞C2BBe1發炎、氧化壓力及凋亡反應之影響。實驗共分為8組,為控制組、LPS組、100、250、500 μg/mL的LBP組及75、50、100 μM的CAP 組。細胞依分組先介入所對應的LBP或CAP劑量後,再以25 μg/mL的LPS與介入物共培養24小時。結果顯示,介入LBP與CAP可降低因 LPS造成升高的介白素-6 (interleukin-6, IL-6) 與介白素-8 (interleukin-8, IL-8) 分泌量、環氧化酶-2 (cyclooxygenase-2, COX-2) 與一氧化碳合成酶 (inducible nitric oxide synthase, iNOS) 蛋白表現量、核轉錄因子-κB (nuclear factor-κB, NF‐κB) 與細胞外訊號調控激酶 (extracellular signal-regulated kinases, ERK) 路徑活化、脂質過氧化物丙二醛 (malondialdehyde, MDA) 濃度及Bax 蛋白表現量。顯示LBP與CAP介入由25 μg/mL的LPS誘導C2BBe1細胞發炎後,可改善發炎、氧化壓力及凋亡反應。
