Abstract
黃病毒為一單股正鏈RNA病毒透過蚊子與蜱於全球進行傳播。多數黃病毒感染案例可導致類感冒症狀,但部分案例則可能出現神經相關病變並診斷為黃病毒腦炎。然而,黃病毒導致神經病變的機制目前尚未被釐清,尤其是登革病毒。過往研究中指出登革病毒能造成神經微小膠細胞之感染並使其細胞移行力異常增加。神經微小膠細胞於不同刺激下細胞表現活化型態對於腦內微環境能扮演多種不同功能。而為了釐清黃病毒腦炎中神經微小膠細胞活化的可能角色。將三種黃病毒分別接種感染於小鼠後,小鼠均會產生神經病變相關病徵並伴隨高死亡率,同時能於小鼠腦內偵測到黃病毒複製與病毒顆粒產生。組織病理學染色發現感染後所造成之神經細胞毒性與異常細胞型態表達於海馬迴區塊為顯著。免疫表現型偵測則揭露神經微小膠細胞活化與周邊免疫細胞於感染後浸潤於小鼠腦部。細胞激素與基因表現型分析進一步確認於黃病毒感染腦部後免疫細胞表現型趨向第一型活化態。與此同時,小鼠腦部中可偵測到腫瘤壞死因子的表現量增加。以藥理阻斷腫瘤壞死因子生成或系統性過度表達腫瘤壞死因子皆確認其異常表現後所參與的神經病變致病角色。本論文亦證實腫瘤壞死因子主要由星形膠質細胞與神經微小膠細胞所分泌。抑制周邊細胞浸潤於腦部不僅能減緩登革病毒感染造成之死亡同時能減少神經微小膠細胞趨向第一型活化態。本篇論文揭露神經微小膠細胞極化反應於黃病毒腦炎中所扮演之作用。
