Abstract
奈米物質在我們的日常生活當中越來越常見,舉凡化妝品、食品、塗料染料或是生活用品中,都可以看到奈米物質的蹤影。然而一旦物質奈米化之後,許多的物化特性和活性皆會有所改變,讓它具有表面積大和尺寸較小的特點,也使得其更容易透過呼吸道、腸胃道和皮膚被人體吸收。但當奈米物質被人體吸收之後,會對人體造成什麼樣的影響仍然不是那麼清楚,所以本篇研究想要深入了解奈米物質對生理上的影響為何。
先前的研究指出,當人類臍靜脈內皮細胞以大小約為40 nm的奈米金處理過後,其細胞通透性會增加,本篇研究則進一步探討了掌管細胞通透性的重要蛋白之一——水通道蛋白 (Aquaporin) 在人類臍靜脈內皮細胞及小鼠腦內皮細胞中的表現。我們發現當人類臍靜脈內皮細胞經過微米金 (630-640 nm) 的處理後,AQP1的蛋白表現量並沒有變化;然而經過奈米金的處理,AQP1的蛋白表現量會隨著奈米金處理的劑量和時間增加而增加,在小鼠腦內皮細胞中也得到了相同的結果。接著我們做了經內皮水通透性試驗,發現隨著奈米金的處理時間增加,兩株血管內皮細胞的水通透性皆會跟著增加。而在小鼠動物模式上,我們以鼻腔吸入的方式給予奈米金溶液,結果顯示小鼠腦中的水分含量會明顯的上升,證實奈米金的確會造成內皮細胞水通透性的增加,也得知奈米金可能會促使小鼠腦部組織之壞死。此外,在AQP1的訊息傳遞路徑上,經過奈米金的短時間處理後,發現AKT及FAK參與在了奈米金對血管內皮細胞之AQP1的調控之中。另外,我們利用免疫螢光染色及免疫沉澱法,發現在人類臍靜脈內皮細胞中,AQP1與Caveolin-1和F-actin之間皆有交互作用。
從目前的實驗結果可以得知,奈米金會透過AKT及FAK促使血管內皮細胞中的AQP1表現量增加,也會使血管內皮細胞之水通透性上升,並在小鼠動物模式中得到驗證;而AQP1與Caveolin-1和F-actin之間的關係仍有待後續實驗探討。
