Abstract
減重方式有許多種,其中飲食控制配合運動介入是最常見的。然而減重飲食對於運動能力的影響卻較少被探討。生酮飲食是一種高脂、低醣及中低蛋白質比例的減重飲食,被認為有助於增進運動員的運動表現。然而其對於肥胖者的影響尚不明確,因此本次研究希望了解生酮飲食在減重期間對於耐力運動能力之影響。C57BL/6J小鼠以高脂飲食誘導肥胖後分為三組:生酮飲食減重組、低脂飲食減重組、以及持續維持高脂飲食陰性對照組,餵食九周。生酮及低脂飲食具有相似的減重效果,但生酮飲食可能透過增加酮體利用能力表現出較好的耐力。為全面的探討飲食造成之蛋白質表現變化,我們利用同重同位素串聯質譜標定之定量蛋白質體學分析肝臟、比目魚肌及腓腸肌之蛋白質組成。其中肝臟及腓腸肌對於飲食影響更為敏感。他們透過改變粒線體電子傳遞練複合物以維持有氧運動中的ATP供應。生酮飲食增加了腓腸肌中以脂肪酸氧化為主的第一型肌纖維組成,增加氧化磷酸化代謝並限制脂肪酸合成。總結而言,我們深入探討了在體重變化中飲食相關的生理適應機制,並解釋了生酮飲食如何增加高脂飲食誘導肥胖小鼠的運動耐力。
