Abstract
過敏是免疫系統對環境刺激的反應,在人類社會中是主要的健康問題。日益嚴重的環境、空氣、工業化國家增加等因素,導致全球過敏人數向上激增。據文獻資料指出: 蛋白質如果與已知的過敏原相比,具有任一6到8個連續氨基酸的相同性或在80個胺基酸內,有大於35%的序列相似,該蛋白質即是潛在的過敏原。而利用此論點,使用機器學習方式進行資料探勘,從巨量的蛋白質序列資料中找到規則,進而,找出過敏原的重要特徵做出預測。


本研究使用了AlgPred、AllerTop、AllerHunter、Allerdictor、Allergenonline 此五個資料庫的資料,將資料區分為訓練組和驗證組,運用SAS Enterprise Miner軟體進行StatExplore、自動類神經網路、決策樹、羅吉斯迴歸等資料探勘技術,找出過敏原的重要特徵,最後再評估多個機器學習的結果做模型比較。

我們發現有些變數在不同的資料組中,會有重複值,而重複的變數較多是帶正電的氨基酸,透過這樣的結果,可以證明蛋白質序列確實有存在著一些重要特徵,導致蛋白質序列為過敏原,也驗證出透過這樣的方法,利用序列相似度可以發現潛在的過敏原。而在模型比較中,在我們最好的AUC有高於90%,而平均的AUC均有85%以上,從而得知機器學習方法是用於蛋白質序列的分析當中。

希望藉由此研究,能讓日後的研究者針對過敏的題材,有更良好的方式進行預測,而也希望能透過結合蛋白質和資料探勘中機器學習的技術,讓此技術多加運用於生物資訊中。
