Abstract
抗癌藥物設計原則,主要針對腫瘤細胞中特定的酶或特定的分子,其機轉通常為抑制其生長或使其自然細胞凋亡。抗癌藥物攻擊癌細胞而不傷害正常細胞,因此,極大地減少了傳統的治療的副作用。
組蛋白去乙醯酶的異常增大是一個普遍存在於許多類型癌症的現象。因此,HDAC 成為抗癌藥物的開發中具有吸引力的標的。最近的研究發現,各種組蛋白去乙醯酶抑制劑能選擇性地抑制腫瘤細胞的生長並誘導細胞凋亡。其中,例如:SAHA 和FK-228,已被美國FDA批准用於治療皮膚T 細胞淋巴瘤。由於HDAC 在癌症治療中具有重大的意義,本論文將著重在針對HDAC 發展有效化合物。
N-的羥基丙烯醯胺和N-羥基乙醯胺對HDAC 抑制是重要的特點,本實驗先前研究證明出部分的aroylindoles 會使活性增強,因此本篇論文決定結合此結構特點,合成出一系列5-aroylindoles 在N1 位置上有N-的羥基丙烯醯胺或N-羥基乙醯胺的化合物,並探討5-aroylindoles 的結構活性的關係。
