Abstract
乳癌是最惡性、最難治療的癌症之一,而且多發生於女性。它在癌症過程中的轉移是極其惡性和致命的。目前對於乳癌轉移尚無良好的治療方法。在這裡我們發現DDX3在乳癌中高表現且與腫瘤幹細胞性和轉移呈正相關。然後我們發現DDX3與粒線體高度相關。我們發現 DDX3 定位於粒線體。透過質譜分析,我們發現DDX3與粒線體分裂的重要蛋白DRP1相互作用,進而影響DRP1的磷酸化。透過影響DRP1的磷酸化,促進粒線體分裂,而粒線體分裂已被證實與許多癌症中的癌症轉移有關。然後,我們在質譜分析中也發現,DDX3與脂肪酸氧化(FAO)相關基因呈正相關,且FAO也被發現會影響腫瘤轉移。因此,當我們添加FAO的誘導劑棕櫚酸 (PA) 時,也發現它可以上調DPR1的磷酸化和粒線體分裂。綜合以上,我們發現在乳癌中,DDX3過度表達,並且DDX3會調節DRP1的磷酸化,進而促進粒線體分裂。粒線體分裂的上調將進一步上調FAO。我們發現DDX3會透過上述途徑影響癌症幹細胞分化 (stemness) 相關基因的表達,進而影響腫瘤球的形成。最後在動物實驗中,我們利用小鼠動物模型,利用小鼠乳癌細胞株4T1培養腫瘤,取得其原位腫瘤以及肺轉移腫瘤。透過腫瘤分離並取其原位腫瘤細胞和肺轉移腫瘤細胞並進行培養,最後透過實驗證實DDX3會在轉移中調控DRP1、FAO以及Stemness表現進而促進惡性程度。綜合以上所述,DDX3可能作為乳癌潛在的治療因子,在腫瘤的發展和轉移中發揮重要作用。
