Abstract
現代醫學中,治療用醣蛋白因其精準的靶向能力及多樣性的醫療應用而顯得極為重要。然而,使用哺乳動物細胞生產的醣蛋白,其醣型通常具有異質性,可能會影響其藥理特性。為了解決這項問題,「化學酵素合成法」成為一項關鍵技術,能製備具有特定醣型的醣蛋白,以提升治療效果。這項技術透過酵素去除原有的多醣鏈,並引入合成的目標醣鏈。
本論文利用Mucor hiemalis 内切糖苷酶(Endo-M)及其突變酶,修飾治療用醣蛋白如單株抗體(adalimumab)及融合蛋白(etanercept)的醣型。我們從sialylglycopeptide中分離出均質的biantennary N型醣鏈(G2S2 N-glycan),並使用兩種外切酶(α-neuraminidase和β-galactosidase)製備去除唾液酸或半乳糖的醣型(G2 N-glycan和G0 N-glycan)。這些半合成多醣進一步用於製備N型醣鏈的高能量中間體,作為精確醣鏈修飾的醣基供體。
研究結果優化了不同N型醣鏈的製備方法,並成功運用了Endo-M及其突變酶實現對單株抗體及融合蛋白的醣型修飾。本研究不僅提供了一種精確控制醣鏈結構的方法,還為未來設計更穩定、更有效的治療用醣蛋白開創了新的可能,推動了新一代生物製劑的發展。
