Abstract
心血管疾病的病程中,血管狹窄是一個重要的過程,內皮細胞的損傷導致細胞激素(cytokine)以及生長因子(growth factors),像是血小板衍生性生長因子(platelet-derived growth factor-BB,PDGF-BB)的釋放,使得原本位在血管中層(media)中的血管平滑肌細胞增生並移動到內層(intima)層,形成較厚的neointima,進而造成血管狹窄(Lee et al., 2006) 。Hinokitiol (2-Hydroxy-4-isopropyl-2,4,6-cycloheptatrien-1-one,β-Thujaplicin),又名扁柏酚,富含於柏科植物中,具有抗發炎、抗腫瘤及殺菌的作用。本論文主要探討hinokitiol抑制血管平滑肌細胞增生的分子機轉。經由MTT實驗,發現hinokitiol可抑制PDGF-BB所刺激的細胞增生。而單獨給予hinokitiol,也會抑制細胞的viability,此外hinokitiol會抑制血清(10% fetal bovine serum,FBS)所刺激的細胞增生。在LDH含量測定的實驗中,發現hinokitiol並不會造成細胞顯著的毒性。而利用BrdU assay可知hinokitiol會抑制PDGF-BB及serum (10% FBS)所刺激的細胞生長。經由西方墨點法發現hinokitiol不會抑制PDGF-BB所刺激的PI3K、Akt、ERK及JAK2的磷酸化。而給予未quiencent的細胞hinokitiol,會增加p21蛋白及p53的磷酸化。此外,在luciferase assay中發現hinokitiol會使p21 promoter 及p53 gene的表現量增加。綜合本篇的實驗結果我們推論hinokitiol經由增加p53的磷酸化而活化其轉錄因子的活性,造成p21蛋白的表現進而抑制細胞週期的進行。Hinokitiol已被證實具有抗發炎的效果(Byeon et al., 2008),本實驗也發現其抑制細胞增生與細胞生長的效果,期許以後hinokitiol在心血管疾病治療上—例如支架上藥料的運用—所扮演的角色。
