Abstract
阿茲海默症 (Alzheimer’s disease, AD) 是神經退化性疾病中最常
見的一種。AD 病徵為神經元外有老化斑塊堆積及神經元內有神經纖
維糾結的現象,分別由β-澱粉蛋白 (amyloid-β, Aβ) 及過度磷酸化的
tau 蛋白質所組成。研究指出,汞可能會造成或加劇AD 的病徵,促
使神經纖維糾結的產生,而體外試驗中也發現,汞會使Aβ 分泌增加
及tau 蛋白質過度磷酸化。另外,汞累積在體內會產生大量的活性氧
屬進而對細胞造成傷害甚至死亡。然而,汞造成細胞死亡途徑仍不清
楚,因此本次實驗將分化之人類神經瘤母細胞 (human neuroblastoma
cells, SH-SY5Y) 與氯化汞共同培養24 小時之後,測其細胞存活率、細
胞內活性氧屬產生量、抗氧化酵素活性、細胞凋亡與壞死及細胞凋亡
下游蛋白質caspase 3 的表現量。隨著氯化汞濃度增加,細胞存活率
隨之降低並呈劑量效應,而細胞內活性氧屬的產生量顯著增加、
glutathione reductase 及glutathione peroxidase 活性顯著降低並呈劑量效應,細胞死亡的部分,細胞凋亡的比例顯著增加、壞死則無顯著差
異,而細胞凋亡下游蛋白質caspase 3 的表現量顯著增加。由結果顯
示,氯化汞造成細胞死亡的原因可能是透過增加細胞內活性氧屬的產
生及降低抗氧化酵素的活性,進而使細胞走向凋亡的途徑。
