Abstract
神經膠質瘤(Glioma)是中樞神經中發生率最高的惡性腫瘤,現行治療方法仍以手術切除、 放射線治療及化學治療為主,化學治療以帝盟多(Temozolomide, TMZ)為腦癌第一線用 藥。由於神經膠質瘤的高復發性(Recurrence),即便在積極治療之下,患者的預後仍不甚 理想。而許多證據指出,腦癌幹細胞的存在與癌細胞產生抗藥性可能是造成腦癌復發的 主要原因。先前文獻中發現麩氨酸接受器次單元 GluR1 與神經膠細胞瘤之疾病進程呈現 正相關。我們的前期研究也證實,腦癌幹細胞及帝盟多抗藥性細胞表面皆會表現大量 GluR1。為了釐清 GluR1 在腦癌中扮演著何種角色,是否是造成腦癌幹細胞或是帝盟多抗 藥性細胞發生的主因,因此我們利用基因轉殖技術讓神經膠質瘤細胞系 U87 細胞大量表 現 GluR1,在 MTT assay 實驗發現 GluR1 過度表現的 U87 細胞相較於原細胞株對於 TMZ 有較高的抗藥性。在反轉錄聚合酶連鎖反應(RT-PCR)以及西方墨點法觀察癌幹細胞指標的 表現,發現 GluR1 過度表現的 U87 細胞,其癌幹細胞指標 CD133、CD15、Sox-2 的 mRNA 表現均增加;CD133、Nanog 的蛋白表現也增加。此外,在傷口癒合(Wound healing)實驗 發現 GluR1 過度表現的 U87 細胞的移行能力高於原細胞株,更進一步的實驗中,篩選可 能影響癌幹細胞特性的細胞傳遞路徑,我們發現 Akt 蛋白在磷酸化以及 Akt 總量皆增 加。本篇研究發現了 GluR1 的過度表現會提升神經膠質瘤細胞的抗藥性、增加神經膠質 瘤細胞的癌幹細胞特性,以及增加細胞移行能力,而有可能透過影響 Akt 蛋白而增加其 癌幹特性。本研究結果顯示,GluR1 有可能是透過 Akt 訊息傳遞路徑造成腦癌復發的關 鍵,在未來具有潛力成為腦癌的惡性指標,也有機會成為未來合併治療並預防腦癌復發 的生物標靶。
