Abstract
睡眠不足與憂鬱症、焦慮症的發生存在雙向關係。Ramelteon (RMT)為臨床上常用的褪黑激素受體致效劑,用於改善失眠患者的睡眠問題。先前研究已指出,n-3多元不飽和脂肪酸(n-3 polyunsaturated fatty acids, n-3 PUFA)具有抗憂鬱的作用;其中,EPA有較顯著的效果。然而,n-3 PUFA是否可以發揮協同效應(synergistic effect),增進褪黑激素受體致效劑的作用,改善憂鬱所產生的失眠症狀,並達到較好的抗憂鬱效果目前還不清楚。因此,本研究欲探討在慢性睡眠剝奪下,預先介入不同n-3 PUFA是否可以提升RMT的作用,改善動物之類憂鬱及類焦慮行為;並從脂質體學的觀點探討RMT與n-3 PUFA介入對腦部類憂鬱及類焦慮行為生物指標的影響。31隻6週齡雄性Wistar大鼠共分為五組,分別為控制組(control, C)、睡眠剝奪組(sleep deprivation, S)、藥物介入組(RMT, SR)、藥物合併EPA介入組(RMT+EPA, SRE)及藥物合併DHA介入組(RMT+DHA, SRD)。研究結果顯示,RMT合併EPA介入在強迫游水試驗中可改善動物之類憂鬱行為;RMT合併DHA介入在高架十字迷宮試驗中可改善動物之類焦慮行為。分析前額皮質蛋白質表現量,RMT合併EPA或DHA介入可顯著提高睡眠剝奪大鼠melatonin receptor 2 (MT2)表現量,且SRE組表現量顯著高於SR組;與S組相比,RMT合併EPA介入顯著提高SIRT1、BDNF及TrkB表現量,合併DHA介入則無此現象。脂質體分析結果發現,只有RMT合併EPA介入可顯著改善所有與行為試驗指標相關之差異代謝物的濃度。綜合上述,RMT合併不同n-3 PUFA可能藉由提高腦部MT2表現量改善動物之類憂鬱及類焦慮行為,而EPA和DHA有不同的效果。
