Abstract
近年來,由於空汙、慢性病引起的肺部疾病增加,加上Covid-19的影響,肺部疾病導致之死亡占比逐年提升,其中晚期肺纖維化比肺癌存活率更低,肺部組織因肺部發炎後造成疤痕組織,導致肺部纖維化、變硬,形成原因不明的纖維化肺泡炎,進而影響肺部功能與日常生活。尋找有效的治療藥物方式,仍是臨床上一大課題。

吡非尼酮 (Pirfenidone, PFD) 為臨床上使用於治療特發性肺纖維化 (Idiopathic pulmonary fibrosis, IPF),但因其所需服用劑量高,且一天服用三次,並有光敏感反應、食慾不振、消化道不適、嗜睡、暈眩及倦怠感等副作用,而讓患者的服藥意願降低,順從性變差;此外呼吸給藥方式可以直接將藥物輸送到肺部,因此改用呼吸給藥方式以提昇PFD肺部作用濃度,可以降低給藥劑量,同時設計奈米劑型,使其具有高效利用率及緩慢釋放效果,來達到改善副作用並提升療效。故本研究將PFD以生物明膠進行包覆,製備成奈米藥物,並採用呼吸給藥方式,遞送到肺部,提升藥物於肺部之濃度,以期達到最佳治療效果。
