Abstract
潰瘍性結腸炎 (ulcerative colitis, UC) 是一種以結腸發炎為特徵的發炎性腸道疾病。儘管存在常規療法,但仍需尋找更安全的替代方案。本研究旨在探討枸杞多醣 (LBP) 與滅活龍根菌OLP-01 (IA OLP-1) 單獨及合併使用時,於脂多醣 (LPS) 誘導之Caco-2腸道上皮細胞模型中的抗發炎潛力。分化後的Caco-2細胞先以LBP與/或IA OLP-1(包含新鮮配製及預混合配方)進行24小時預處理,隨後以相同介入物合併10 µg/mL LPS進行24小時共同處理。細胞激素分泌量以ELISA法測定,關鍵蛋白則以西方墨點法分析。結果顯示,LPS顯著增加了IL-6與IL-8的分泌。在單獨介入組中,IA OLP-1具劑量依賴性地降低了IL-6並顯著降低IL-8;LBP則顯著降低IL-8,並對IL-6呈現降低趨勢。在合併介入組中,新鮮配製及預混合24小時的LBP/IA OLP-1顯著減弱了IL-6的表現,但預混合72小時的配方則無效。值得注意的是,所有LBP/IA OLP-1的組合均未能有效降低IL-8。西方墨點法分析顯示,LPS或任何介入處理均未對COX-2、TLR4、p-IκBα/IκBα比值或緊密連接蛋白 (ZO-1, claudin-1, occludin) 的蛋白質表現量造成顯著改變。總結而言,IA OLP-1、LBP及特定的短期組合在體外模型中展現了降低促發炎細胞激素分泌的潛力。合併使用的複雜結果以及對所測蛋白路徑的缺乏影響,表明其抗發炎作用可能由替代機制所介導,值得進一步探討。
