Abstract
在各種腦神經疾病,例如中風後腦損傷或多發性硬化症、阿茲海默症、帕金森氏症、人類免疫缺乏病毒相關失憶症等神經退化性疾病中,神經膠細胞的活化及發炎反應扮演了疾病發展重要的角色。活化的神經膠細胞所分泌的各種前發炎細胞激素及神經致毒媒介會導致神經細胞的死亡,藉由抑制神經膠細胞的過度活化可以達到減緩神經退化性疾病的發展。本研究想探討第八型組織蛋白去乙醯酶的抑制劑HHJ-16是否會透過抑制小膠質細胞及星狀膠細胞的活化而減少腦部發炎所誘發的損傷。在脂多醣體誘導腦部發炎的小鼠中,HHJ-16能顯著降低MMP-9的活性及COX-2、iNOS的表現量。由免疫螢光染色結果顯示腦部神經小膠質細胞與星狀膠細胞的增生數量也有減少的趨勢。此外,HHJ-16同樣能減少小膠質細胞BV-2活化後所釋放的COX-2與iNOS的表現量且抑制BV-2活化後之增生,p-STAT1與p-AKT表現量下降暗示HHJ-16降低發炎反應是透過調控STAT1與AKT之路徑。因此本研究推論藥物HHJ-16可能藉由調控神經膠細胞活性達到抗發炎及神經保護之特性。
