Abstract
神經系統的發展需要許多神經迴路的精準連結,神經元生長椎經過特定訊號引導,精確到達正確的目標。這些特定訊號透過外在環境的配體以及軸突細胞表面之引導受體共同表達。截至目前為止,僅發現少量的軸突引導受體及其配體,這表示配體及其受體之間有多種相互作用模式得以擴大其用途。我們已成功地利用脊髓外側運動神經柱(lateral motor column, LMC)的運動神經元生長至肢體肌肉組織的兩個主要路徑選擇,執行相關的研究。過去所作的研究已經發現,在LMC中需要一些特定物質去幫助它作出這兩個路徑的選擇,其中包含了Eph酪氨酸激酶感受體的成員和他們的ephrin配體之間的交互作用。最近的研究指出,星狀膠質細胞中的轉錄因子Specificity protein 1(Sp1) 引導神經生長及突觸發生.因此在LMC運動神經元生長中,Sp1對於其路徑選擇,可能扮演重要的角色。為了進一步瞭解Eph酪氨酸激酶感受體及其ephrin配體之調節機制,我們假設Sp1調控LMC神經元中ephrin之表達,其作用是讓LMC軸突能正確地生長到肢體肌肉。首先我們經由原位雜合技術確認Sp1在小鼠胚胎及雞胚胎LMC運動神經元的表達,再來利用雞胚胎電穿孔技術,執行過度及抑制Sp1的表達實驗以確認Sp1對於軸突選擇的影響。並且經過RNA-Seq 的分析去了解Sp1可能調節與ephrin相關的基因。接下來,我們經由離體實驗探討 Sp1 在LMC軸突選擇過程中,是否具有調節Eph訊息之傳遞,以及Sp1 之共同激活因子P300及CBP是否共同參與調節。最後利用Sp1缺乏的基因小鼠,觀察成體後的運動功能表現。藉由以上研究結果,我們發現:
1. 在引導脊髓運動神經生長到肢體肌肉的過程中,Sp1扮演重要角色。
2. Sp1連同其共同激活因子P300及CBP,在LMC軸突引導的過程中,作為調節Eph訊息的重要物質。
3. 確定Sp1突變造成的軸突引導錯誤,會對成年時期的運動功能有所影響。
本篇文章的結論是,Sp1 作為LMC 神經元的的關鍵轉錄因子,在調控ephrin表達及ephrin/Eph 介導的軸突引導中發揮關鍵作用,並提供了神經迴路形成的分子機制新見解。
