Abstract
Doxorubicin為治療多種癌症常見的化學治療藥物,然而抗藥性的產生往往是造成化學治療效果不佳的主要原因之一,因此,如何減少抗藥性與藥物的使用量,並減少藥物副作用,以達到更好的治療效果,以及開發新的治療物質成為重要的課題。子宮惡性肉瘤為罕見的婦科癌症,其有時無明顯症狀,惡性程度極高,預後差且死亡率高,診斷上不易與子宮肌瘤區分。異甘草素 (isoliquiritigenin, ISL) 是源自於甘草根中的類黃酮物質,目前已有多項文獻指出其具有抗癌的作用。因此本次研究的目的在於探討異甘草素是否能夠抑制人類子宮惡性肉瘤細胞與具多重抗藥性的人類子宮惡性肉瘤細胞,及相關的死亡機制。本研究主要使用人類子宮惡性肉瘤細胞MES-SA和具多重抗藥性細胞MES-SA/Dx5與MES-SA/Dx5-R等進行研究,實驗結果發現,異甘草素可以選擇性抑制癌細胞MES-SA、MES-SA/Dx5與MES-SA/Dx5-R的生長,以特定濃度比例合併異甘草素和doxorubicin處理對細胞生長抑制效果具有相乘作用 (synergistic effects),並增加細胞週期G2/M和subG1的比例,由流式細胞儀分析發現細胞凋亡的比例顯著增加,且會促進細胞凋亡相關蛋白cleaved PARP、cleaved caspase-7、bax/bcl-2比例及自噬作用相關蛋白LC3BⅡ的表現,抑制磷酸化mTOR之表現;異甘草素亦會抑制人類子宮惡性肉瘤細胞的移行能力。因此,異甘草素對於抑制人類子宮惡性肉瘤細胞具有多重之效果。
