Abstract
非酒精性脂肪肝疾病 (non-alcoholic fatty liver disease, NAFLD)為目前全球最常見的慢性肝臟疾病之一。在誘導NAFLD的動物模式中,cafeteria (CAF)飲食為在動物基礎飼料中混入人類食用之零食及加工製品,具有高脂肪、高糖、高鹽、富含飽和脂肪和膽固醇之特性。兒茶素及咖啡因為抹茶主要成分,被認為可有效抑制脂質堆積、抗發炎及促進腸道和大腦健康等作用。然而,直接投予抹茶是否能藉由改變腸道菌相進而改善大腦認知功能,此一機制仍需更多研究進行佐證。故本次實驗以腸道健康及大腦行為之觀點探討抹茶對於cafeteria飲食誘導大鼠NAFLD的影響。本實驗為期16週,使用40隻4週齡雄性Wistar大鼠,以cafeteria飲食誘導NAFLD,並於飼料中介入抹茶粉。實驗於適應期後將大鼠分為正常飲食組 (control, C)、正常飲食 + 0.2%抹茶粉組 (C + 0.2%M)、CAF飲食組及CAF + 0.2%M組,於實驗期第16週進行犧牲。本次研究使用之cafeteria 飲食可誘導肥胖、血漿triglyceride (TG)濃度上升、胰島素阻抗、肝臟損傷及大腦發炎指標上升,並可能與腸道菌相多樣性下降、腸道緊密連接蛋白zonula occludens-1 (ZO-1)和occludin濃度降低及toll-like receptor (TLR4)/myeloid differentiation primary response 88 (MyD88)路徑活化相關。而cafeteria飲食中添加0.2%之抹茶粉可降低血漿TG濃度、改善肝損傷及增加腸道ZO-1濃度,說明抹茶對於改善NAFLD具有一定的潛力。
