Abstract
肥胖是目前各國主要公共衛生議題題之一,女性由於荷爾蒙影響容易造成肥胖,因此應多加注意育齡女性的體重控制,以避免影響停經後的生活品質。本研究透過給予停經前體重正常之雌性大鼠褪黑激素及不同比例脂肪酸之飲食,探討對於脂質代謝和身體組成的影響。實驗由32隻8週齡雌性Sprague Dawley大鼠模擬停經前體重正常之女性,隨機分為控制組(Control, C)、褪黑激素組(Melatonin, M)、特殊比例油脂飲食組(Experimental oil diet, E)及褪黑激素合併特殊比例油脂飲食組(Melatonin + experimental oil diet, ME),特殊比例油脂飲食為PUFA / SFA = 5,MUFA = 60%之飲食,褪黑激素以管餵的方式於每天17:00 – 19:00介入50 mg/kg/day,共持續8週。實驗結果顯示,M組有提升血清 adiponectin 濃度、降低 leptin 濃度的趨勢,並促進與脂質氧化相關基因(如 CPT-1 與 HSL)的 mRNA 表現。E組則可降低血清 leptin 及肝臟中 TC、TG 濃度,抑制脂質合成相關基因表現,同時提升PPARα的表現,且CPT-1及HSL的mRNA表現量達顯著上升。ME組整合兩者介入的效應,可降低血清FFA濃度,使肝臟中TC、TG、ACC、SREBP-1c及脂質組織中LPL的mRNA表現有下降的趨勢,同時使血清adiponectin濃度、肝臟CPT-1、脂肪組織中HSL的mRNA表現具有上升的趨勢,且肝臟PPARα的mRNA表現亦達顯著提升。顯示兩者合併介入可促進脂質分解、抑制脂質合成,並且有助預防育齡女性肥胖,進而降低停經後代謝異常風險及提升生活品質。
