Abstract
DNA的甲基化大多發生於啟動子的區域,在腫瘤的生長過程中是一個很常見的現象。藉由DNA甲基轉移酶在CpG島 (CpG island)上的高度甲基化作用能夠完全的關閉抑癌基因的表現。脾酪胺酸蛋白激酶 (Syk) 是一種非受體型的酪胺酸蛋白激酶 (non-receptor tyrosine kinase),在不同的腫瘤細胞中可扮演抑癌蛋白或是促進癌症生長的角色。已經有研究指出Syk基因在乳癌、胃癌、黑色素癌、肺癌以及大腸癌中有高度甲基化的現象,而目前Syk基因甲基化與p53之間的關係仍然不清楚。在大腸癌HCT116細胞株中我們發現,Syk的蛋白質與mRNA表現量在HCT116 p53-/-細胞中都顯著地低於HCT116 WT細胞。不僅如此,在HCT116 WT中使用p53的抑制劑PFT-α與si-p53都可以降低Syk蛋白質與mRNA的表現,而在HCT116 p53-/-細胞中使用DNMT抑制劑 5-Aza-2'-dC可以增加Syk蛋白質與mRNA的表現。另外在HCT116 p53-/-的細胞中,其DNMT的表現量是比HCT116 WT來的高,而且PFT-α在HCT116 WT細胞中不僅可以增加Syk基因啟動子(promoter) 的甲基化現象,還可以增加DNMT1的蛋白質與mRNA的表現,這些結果顯示p53可以負向調控DNMT1的基因表現,進而參與Syk基因甲基化的作用。在另外兩株惡性的肺癌細胞株A549與PC9中 (其各自帶有WT p53與gain of function p53),我們也發現PFT-α在這兩株細胞中可以顯著降低Syk mRNA與蛋白質表現,然而只有在A549細胞株有出現Syk基因甲基化增加的現象。在Annexin V-PI的染色實驗與MTT的細胞活性實驗中,我們發現Syk抑制劑R406能夠降低HCT116 WT、A549以及PC9細胞的活性以及增加細胞的凋亡。總結來說,p53能夠藉由抑制DNMT1基因的轉錄來影響Syk的基因甲基化程度,而此調節Syk甲基化的作用會隨不同癌細胞的狀態改變,通常在低p53表現的癌細胞中Syk甲基化的調節作用較明顯。因此我們認為瞭解Syk的表觀基因遺傳調節機制及其腫瘤生物作用後,希望可以對將來癌症治療提供一個新的方向。
