Abstract
視網膜色素上皮細胞 (RPE) 是視網膜中重要的提供營養、維護和代謝之細胞,也是血視網膜屏障 (BRB) 的重要組成部分。在糖尿病視網膜病變與老化黃斑部病變等疾病上,視網膜代謝及其發炎異常導致RPE功能受損是相當重要的因素。許多文獻顯示,老年性與糖尿病視網膜病變病患之眼內沉積物甲基乙二醛 (Methylglyoxal, MGO) 也是造成 RPE 損傷之因素。本研究主要探討兩種真菌天然物WCS1-32-6和4AAQ是否能緩解MGO以及 LPS 刺激 BV2 發炎介質誘導 ARPE-19 之細胞發炎與損傷反應。實驗結果顯示,代謝毒物 MGO 在特定時間下,明顯造成ARPE-19細胞存活率隨濃度梯度下降。在凋亡蛋白的探討中,可發現 MGO 能誘導PARP-1蛋白表現的減少與Cleaved-Caspase 3的增加。WCS1-32-6可顯著減少 MGO 誘導 ARPE-19 細胞的損傷死亡。同樣地,WCS1-32-6可明顯減少NaIO3 誘導ARPE-19 氧化損傷之死亡。另一方面,在TNF-α刺激下或以LPS刺激BV2之培養基共同培養後,WCS1-32-6對於上述刺激下ARPE-19的MMP-9活性與蛋白質表現量具有顯著的抑制作用,但4AAQ無顯著抑制作用。訊息路徑中,可發現LPS刺激BV2之培養基共同培養後誘導ARPE-19細胞MMP-9活性表現主要與NF-κB、JNK、ERK以及STAT3有關。 WCS1-32-6 對於 LPS 直接活化 BV2 產生發炎介質 (TNF-α及COX-2) 並無顯著抑制作用,但 4AAQ 可以顯著抑制 LPS 誘導之 BV2 的 COX-2蛋白質表現。此外,針對不同發炎細胞 THP-1,4AAQ顯著抑制 LPS 誘導之 MMP-9 生成及釋放。在 LPS 活化之 THP-1 細胞中, 4AAQ可呈現濃度梯度的抑制STAT3 (Y705) 之磷酸化。根據本研究的嶄新發現,WCS1-32-6與4AAQ個別具有不同的保護RPE與抗發炎之作用,未來將進一步探討NaIO3在各種RPE損傷模式中之保護效果與運用。
