Abstract
本次研究的目的為探討給予高血糖小鼠管灌不同劑量之褪黑激素對於其血糖恆定、抗氧化能力及脂肪激素之影響。實驗將50隻ICR小鼠分為控制組 (control group, C) 與糖尿病組,糖尿病組又分為 (diabetic group, D)、低劑量 (10 mg/kg, L)、中 (20 mg/kg, M) 及高劑量組 (50 mg/kg, H)。誘發高血糖之後,每日於17:00-19:00間給予各組不同劑量之褪黑激素,六週介入期結束後犧牲。分析肝臟與血液脂質、氧化壓力指標、瘦體素及脂聯素。結果顯示,D組出現高血糖、氧化壓力增加、血脂異常及脂聯素低下等與糖尿病相關的症狀;介入褪黑激素之後,雖介入組血糖值沒有降低,但H組血糖上升幅度顯著較D組低且胰島素濃度顯著回升,血漿及肝中三酸甘油酯與膽固醇濃度皆顯著較D組為低。介入組肝臟MDA濃度顯著降低,其中H組肝臟中抗氧化酵素活性顯著回升。介入組血漿脂聯素顯著增加,瘦體素與瘦體素/脂聯素比值顯著下降。本次研究發現在NA/STZ誘發高血糖模式下,給予管灌六週50 mg/kg BW褪黑激素可減少氧化壓力、降低瘦體素/脂聯素比值、減緩血糖上升幅度及改善葡萄糖耐受性。
