Abstract
本篇研究目的為探討高不飽和脂肪酸與飽和脂肪酸比值 (P/S ratio) 搭配不同含量的單元不飽和脂肪酸對3T3-L1脂肪細胞脂質生合成及脂質相關代謝之作用機轉。3T3-L1前脂肪細胞先培養於含dexamethasone、3-isobutyl-1-methylxanthine及insulin的Dulbecco’s modified eagle medium (DMEM) 培養液中,再以含insulin的培養液培養使其分化為成熟脂肪細胞後,分別用體積比1: 10: 5及1: 2.5: 5的方式混合棕櫚酸、油酸及亞麻油酸培養脂肪細胞 (相等於P/S ratio=5; MUFA 60%,簡稱M60及P/S ratio=5; MUFA 30%,簡稱 M30) 。經過72小時後,以油紅染色分析脂肪酸對細胞內脂質堆積程度的影響,並分析細胞內三酸甘油酯 (TG) 的含量; 同時分析培養基中甘油的釋出量以及利用real-time PCR的方法分析脂質代謝相關基因peroxisomal proliferator-activated receptor γ (PPAR γ) 、leptin、adiponectin、glucose transporter-4 (GLUT-4) 、hormone-sensitive lipase (HSL) 及lipoprotein lipase (LPL) 的表現。結果發現,10 μM低劑量的M60組 (M60-10) 與同劑量的M30組 (M30-10) 相比,可顯著降低細胞內TG含量以及PPAR γ、LPL、leptin、adiponectin、GLUT-4 mRNA的表現量,同時能增加脂解作用相關酵素HSL mRNA的表現,進而達到降低脂肪細胞三酸甘油脂蓄積的效果。
