Abstract
以全球的流行病學來看,大腸直腸癌的發生率在男性排名第三,女性排名第二,大腸直腸癌主要分零期到第四期,儘管目前的治療已有大幅進步,但對於晚期轉移性癌症的病人五年存活率仍低,可見大腸直腸癌對於人類健康的威脅極大,因此探討關於大腸直腸癌進程的分子機轉是件重要的事。

HMGA2含有3個AT hook domains結構以調控下游目標,促進或抑制基因的表現。一般來說,HMGA2只會在人類早期發育表現,但文獻指出若在成人中過度表現,即和良性惡性腫瘤及癌症突變有關,儘管目前已有許多文獻證實HMGA2在許多癌症中皆會大量表現,但極少探討HMGA2和下游基因的關聯性,因此本研究目的是要探討HMGA2和下游目標對於大腸直腸癌進程之調控。

本研究使用定量反轉錄聚合酶鏈鎖反應和西方墨點法來分別偵測細胞中基因和蛋白質的表現量,藉由luciferase reporter assay證實HMGA2是否調控TGFα啟動子的活性,HMGA2和TGFα對於大腸直腸癌細胞增殖、移動和侵襲的影響則由細胞增殖、細胞群落、傷口癒合、細胞遷移、細胞侵入實驗來探討。

由實驗結果顯示,在DLD-1大腸直腸癌細胞中,當HMGA2的基因和蛋白質表現量上升,TGFα的表現量也上升,且luciferase reporter assay證實HMGA2會活化TGFα promoter,在大腸直腸癌細胞的功能性實驗中,發現當HMGA2及TGFα表現量上升,皆會促進細胞增生、造成細胞遷移和侵入速度增快。

整體而言,研究研究結果指出在大腸直腸癌細胞中,HMGA2可能藉藉由調控TGFα表現來促進細胞生長及增加細胞移動和侵襲能力,藉此加速大腸直腸癌之進程,因此HMGA2和它的下游目標TGFα可作為未來臨床上預測大腸直腸癌進程的生物標記。
