Abstract
研究背景:
牙植體已經廣為使用於替換單顆或多顆缺牙,但植牙後骨修復時間需要3至5個月,為了縮減骨癒合時間,我們改善植體的骨整合性,骨整合性與骨傳導性、骨誘導性息息相關,也是骨再生至關重要的部分。Ti6Al4V由於有優異的機械性質及骨相容性, 再經由塗覆磷酸鈣塗層的表面改質可增加其骨傳導性,故常被選作為骨植體。加載生長因子可以加速成骨細胞分化(骨誘導性),而常使用的生長因子為BMP-2、IGF-1及TGF-1。此外,利用電化學沉積法披覆仿生層以及加載生長因子是更加穩定且有效率的。
研究目的:
在這項研究中,我們利用電化學沉積法開發了具骨傳導性的多孔鈦(Ti)與磷酸鈣(CaP)塗層加載骨誘導性之生長因子-骨形態發生蛋白(BMP),以研究其在植體上的特性。
材料與方法:
利用三種不同製程之電化學沉積法製備磷酸鈣塗層於Ti6Al4V表面,並將生長因子與磷酸鈣共沉積於鈦基板。首先找出披覆磷酸鈣的最佳電位,披覆完成後再以掃描是電子顯微鏡觀察塗層形貌,並以alpha step測得其厚度,以及XRD分析其化學組成。以電化學沉積法將BMP-2加載於Ti-CaP上(Ti-CaP-BMP),並將其進泡於模擬體液(SBF)評估其載藥及釋放效率,以及與MG-63細胞共培養之細胞增植性、細胞分化性。

結果
磷酸鈣成功得沉積於Ti6Al4V表面,結果表明,ECD在-0.4 V到-1.6 V的電位下沉積表現出較好的CaP性質。SEM觀察顯示出CaP均勻的沉積並具有孔洞結構;透過XRD分析表明Ti-CaP的磷酸鈣組成為磷酸氫鈣(DCP),並且CaP披覆於鈦合金上有良好的結晶度。藉由浸泡模擬體液,Ti-CaP樣品皆能促使磷酸鈣生長。此外,由BET測試得出Ti-1.6呈現較大的孔洞尺寸及總孔洞體積,並從表面接觸角發現電壓越高其表面粗糙度越高。然而,於臨床上生長因子的遞送應用在骨修復是相當重要的,Ti-0.8V-BMP能夠長時間釋放,並擁有最好的細胞增值性及分化性。
結論:
我們成功利用電化學沉積法在Ti6Al4V樣品上沉積磷酸鈣多孔塗層,隨著ECD所使用的電壓上升,將增加圖層的厚度、粗糙度以及孔洞大小;從XRD及SEM圖中得到,磷酸鈣再浸泡模擬體液中會產生礦化。BMP-2藉由ECD共沉積於Ti-CaP上,並有長期的效應,而Ti-0.8V-BMP具有最好的細胞活性、細胞增植性及細胞分化性。
