Abstract
髓母細胞瘤(MB)是兒童最常見的惡性腦腫瘤。常規化療藥物有長春新鹼 (vincristine)、順鉑(cisplatin)和依托泊苷(etoposide)被用於治療髓母細胞瘤的患 者。研究顯示,長春新鹼和順鉑僅在治療初期有效,但腫瘤細胞隨後會對化療藥物產 生抗藥性,阻礙治療效果。過去的研究已經證明,熱休克蛋白(HSPs)在多種癌症中 表達增加,並且可能與癌細胞的增殖、分化、轉移和死亡有關。此外,先前的研究也表明,熱休克蛋白在某些抗藥性癌細胞,如結直腸癌、膀胱癌、卵巢癌和某些癌症幹 細胞(CSCs)中扮演重要的角色。因此,我們假設熱休克蛋白可能影響髓母細胞瘤的癌幹性和化療抗藥性。通過西方點墨法,我們證明了在 CD133+ DAOY 和 CD133+ D283MED 髓母細胞瘤癌幹細胞(MBSCs)中,熱休克蛋白70(HSP70)表達增加。 為了探討 HSP70 在化療抗藥性和癌幹性中的作用,我們使用siRNA 技術在 DAOY 和 D283MED 癌幹細胞中敲低 HSP70 的表達,結果顯示 HSP70 表達減少,並伴隨 MB 癌幹細胞表面標誌物、存活率和球體形成能力的下降。此外,暴露於HSP70 抑制劑VER155008的 MB癌幹細胞也顯示出 HSP70、癌幹細胞標誌物、增殖和球體形 成能力的顯著減少,這與使用HSP70 敲低的結果一致。為了評估聯合化療藥物和VER155008 對 MB 癌幹細胞的治療效果,將細胞暴露於 VER155008 結合順鉑或長春 新鹼,結果顯示與僅用順鉑或長春新鹼治療的細胞相比,存活率顯著降低,凋亡細胞 數增加。總結來說,我們的數據表明HSP70是治療抗藥性 MB 的一個潛在良好的靶向分子。
