Abstract
甜菊糖苷 (Stevioside) 是由草本植物甜菊所萃取出的一種成分,因其色澤純白、無異味,常被用來作為甜味劑,近來有研究發現,甜菊糖苷可以用來改善第二型糖尿病的症狀。AMP-activated protein kinase (AMPK) 是一個感應細胞內能量代謝平衡的關鍵調控者,磷酸化的AMPK可以減少脂肪合成和增加脂肪酸的代謝,因此,AMPK已被廣泛的認為是治療第二型糖尿病的分子標靶。我們篩選了數種由甜菊糖苷修飾後的化合物,發現編號HSU-STD-012的化合物可以有效的刺激C2C12細胞中AMPK磷酸化,活化的AMPK也抑制其下游acetyl CoA carboxylase (ACC) 的活性,提高carnitine palmitoyl transferase 1 (CPT1) 的表現而促進脂肪酸進行β氧化作用。此外,我們也發現HSU-STD-012可以透過AMPK-p38路徑增加細胞對葡萄糖攝取量。在動物實驗中,我們使用nicotinamide及streptozotocin (STZ) 誘發小鼠產生第二型糖尿病後,投予藥物治療,發現HSU-STD-012可改善糖尿病鼠的葡萄糖耐受度,且降低體重和內臟脂肪堆積。根據以上結果,HSU-STD-012可以透過AMPK傳遞路徑調控醣類及脂肪代謝,在未來可能可以做為一個新穎的治療代謝症候群及糖尿病的藥物。
