Abstract
凝血酶 (Thrombin)在肺部發炎及肺部疾病中扮演相當重要的角色。先前的研究發現,thrombin可以刺激肺部上皮細胞表現interleukin-8 (IL-8/CXCL8),也證實了IL-8/CXCL8 在呼吸道發炎疾病中扮演關鍵的角色。我們先前的研究發現,在人類肺部上皮細胞 (A549 細胞)中,thrombin會經由Protein Kinase C (PKC) α/c-Src、Rac1/PI3K/Akt與IкB Kinase α/β (IKK α/β)/NF-кB路徑誘導IL-8/CXCL8的表現。本論文將深入探討mammalian target of rapamycin (mTOR)、p70S6K、p300與NF-кB等訊息傳遞蛋白在thrombin誘導人類肺部上皮細胞表現IL-8/CXCL8所扮演的角色。首先我們使用rapamycin (mTOR抑制劑)、p70S6K siRNA與p300 siRNA可以抑制thrombin誘導IL-8/CXCL8表現。我們也發現thrombin可依時間相關性誘導mTOR Ser2448、p70S6K Thr389、p300 Ser1834磷酸化。使用rapamycin可以抑制thrombin誘導的p70S6K磷酸化。使用rapamycin及p70S6K siRNA可以抑制thrombin誘導的p300磷酸化。此外,使用dominant negative mutant of Akt (AktDN) 可以抑制thrombin誘導mTOR、p70S6K與p300磷酸化。Thrombin可依時間相關性誘導磷酸化的p70S6K從細胞質轉位進入細胞核中,並促使p70S6K、p300與p65形成複合物。Thrombin可依時間相關性誘導p65 Lys310乙醯化,並促使p70S6K、p300與p65複合物結合於IL-8/CXCL8 promoter上。使用rapamycin及p70S6K siRNA可以抑制thrombin誘導的кB-luciferase activity。綜合以上所有研究結果,在人類肺部上皮細胞中,thrombin會經由活化mTOR/p70S6K/p300訊息傳遞路徑誘導NF-кB的活化,啟動IL-8/CXCL8 promoter並促使IL-8/CXCL8表現。
