Abstract
敗血症所導致的多重器官衰竭目前仍是加護病房照護最主要的死因。敗血症是一種全身性的發炎反應,主要是由葛蘭氏陰性菌上的內毒素 (Lipopolysacchride)所引發的,又稱全身性炎性反應症候群 (SIRS)。有多種組織蛋白去乙醯酶抑制劑已經被合成,目現正在進行抗腫瘤藥物的臨床試驗,此類作用成分也具有抗發炎的效果。因此本研究室藉由LPS誘發敗血症的動物模式,去探討組織蛋白去乙醯酶抑制劑對敗血症的影響,觀察器官損傷的程度、發炎指數及MMP活性的變化來評估藥物作用。
本研究探討新型組織蛋白去乙醯酶抑制劑PBHA衍生物AN對於LPS誘發小鼠敗血症的影響。由實驗結果顯示,AN在LPS誘發敗血症的小鼠上,觀察到對敗血症後體溫的下降有明顯改善,對血液中異常增高的TNF-α和IL-6有降低效果。特別地,在肺部組織發炎蛋白iNOS的表現具有明顯抑制效果,肺部組織的發炎細胞浸潤有改善。綜合以上實驗結果,本研究顯示AN可藉由抑制發炎介質的產生,達到治療敗血症的作用,期望以此發現做為未來抗發炎及敗血症藥物開發與治療的新策略與依據。
