Abstract
研究背景
幹細胞治療的研究帶來現代醫學治療的新契機,在幹細胞的骨組織工程中,骨髓幹細胞(BMSC)是最常被拿來做研究的,因為它具有大量的細胞且容易分離,然而採集骨髓幹細胞必須侵入性採集,這是做細胞移植的一個限制。因此,要找到代替骨髓幹細胞並且促進骨分化,提高骨再生的成功率是關鍵的問題。人類臍帶間葉幹細胞(WJMSCs)是有潛力的,可代替骨髓幹細胞,WJMSCs取得容易且分化能力很強。無患子屬於無患子科無患子屬,有研究指出無患子具有生物效應,可治療癌症、抗發炎、抗心血管疾病……等,然而無患子油對WJMSCs是否有促進骨癒合效果仍不清楚。

目的
本實驗的目的探討添加無患子油對人類臍帶間葉幹細胞(WJMSCs)增生與分化的影響。

材料與方法
本研究實驗中首先添加不同濃度的DMSO作為乳化劑,進行WJMSCs的細胞活性檢測,並測量DMSO對無患子油的最高溶解度,再分析無患子油對WJMSCs之影響,以流式細胞儀檢測WJMSCs添加無患子油後的表面特徵蛋白。接著進行添加無患子油對WJMSCs骨分化影響之實驗,並探討其訊息傳遞路徑是否與MAPK pathway相關。為了瞭解無患子油中影響骨分化的有效成分,我們將細胞培養於含有骨分化的 培養基中,實驗時以Alizarin Red S solution染色並進行鈣化沉積的定量分析,而導致WJMSC進行骨分化之訊息分子機轉,則以即時定量聚合酶連鎖反應(Real-time PCR)來確認。

結果
從結果發現,當以不同濃度的DMSO作為乳化劑進行細胞活性實驗時,由結果可得知0.2% 的DMSO不會傷害細胞,而不同比例的無患子油與DMSO的最高溶解度為2:5。在DMSO與無患子油的細胞活性與骨分化的實驗中,分別以細胞活性MTT與Alizarin Red S solution染色與定量後,結果發現添加無患子油可促進細胞增生與分化,添加抑制劑則都有抑制骨分化的效果。經由成分分析發現油酸與棕櫚酸是無患子油促進骨分化的重要成分。ALP與OCN在添加無患子油表現量都增加。

結論
從結果得知添加無患子油有促進細胞增生與導致骨分化的效果,其中油酸與棕櫚酸是無患子油促進骨分化的重要成分。此外本研究發現添加無患子油導致骨分化與extracellular signal-regulated kinase (ERKs)、c-Jun N-terminal kinase (JNKs)、p38 MAPK訊息傳導路徑有關。而WJMSC的ALP與OCN基因在添加無患子油後表現量都增加,因此添加無患子油在臨床上有增加骨癒合的趨勢。
