Abstract
心血管疾病為近年台灣十大死因之一,心血管疾病的預防是一個重要的課題。心血管疾病中又以冠狀動脈疾病(coronary artery disease,簡稱CAD)最為常見。導致CAD的危險因子眾多,如高血壓,高血脂,糖尿病等等。也有許多證據顯示,遺傳因子在CAD的發生上扮演一個重要的角色。本研究徵求150位民眾(其中實驗組100位,對照組50位)為樣本,針對目前已知在染色體上和CAD可能有關的四組相關基因的多型性(polymorphisms),以統計的方式分析其中致病的相關性。
實驗中四組和CAD相關的基因分別是mutation of connexin37 (Cx37 P303S),one nucleotide insertion promoter of matrix metalloproteinase-3 (MMP3 5A -1171 6A),mutation of paraoxonase-1 (PON1 Q192R),mutation of paraoxonase-2 (PON2 C311S),我們利用PCR-RFLP (polymerase chain reaction-restriction fragment length polymorphism)的方法分析此四組相關基因的多型性變化。
結果顯示,在這四組相關基因的多型性中Cx37、PON1與PON2會增加罹患CAD的危險性,但MMP3沒有明顯的證據增加CAD的危險。台灣CAD病人可能有不同於這幾組的基因多型性,繼續尋找可能的相關突變或其他的候選基因相是未來的研究重點。
