Abstract
在後天免疫系統中樹突細胞主要角色是參與T細胞的調控,樹突細胞經過抗原刺激後會促進T細胞的活化及naïve CD4+ 細胞的分化。樹突細胞表面的Notch ligands能活化T細胞上的Notch訊息傳遞,Notch訊息與細胞週期、分化及凋亡間關係密切,而且在哺乳類動物上Notch訊息被認為對於免疫細胞分化息息相關。研究顯示經由樹突細胞上Delta類型的Notch ligands與T細胞上的Notch receptor接觸,能促使T細胞偏向Th1細胞分化,故在本實驗中,我們構築起帶有小鼠Delta1基因的腺病毒載體,並藉由此帶有Delta1基因的腺病毒來感染樹突狀細胞,使樹突狀細胞表現出Delta1蛋白,從而觀察此過量表現Delta1的樹突細胞所具有的活化與成熟的特性,以及此種樹突狀細胞對於T細胞活化與分化所造成的影響。首先在使用Ad-Delta1腺病毒感染後的樹突細胞的確能增加Delta1表面膜蛋白的表現,接下來在in vitro情況下,觀察到經OVA刺激下有Ad-Delta1(Ad-DLL1)感染的樹突細胞(DC/Ad-DLL1)在活化與成熟上有所上升,並分泌較多促進Th1生成的IL-12細胞因子,並檢測到此種DC/Ad-DLL1能有效促進活化的T細胞分化成Th1型細胞並且產生更高量的IFN-γ。接著我們將這群經過Ad-Delta1修飾後的樹突狀細胞送入OVA誘導的過敏性氣喘老鼠體內進行治療,從DC/Ad-DLL1來治療氣喘小鼠的模式中可以觀察到,DC/Ad-DLL1治療組別中OVA專一性IgG2a含量增加,並降低OVA專一性IgE及IgG1的生成。此外,DC/Ad-DLL1可以引起肺部的Th1免疫反應並有效抑制發炎細胞浸潤以及Th2免疫反應的發生,改善氣管過度收縮反應吸肺部發炎現象。在脾臟細胞免疫反應中DC/Ad-DLL1組別能觀察到相對高量的IFN-γ產生,表示免疫反應偏向Th1族群方向,且減低了IL-4,IL-5和IL-13等Th2免疫反應相關細胞激素產生。綜合以上結果,我們認為Delta1表現的樹突細胞能有效誘導Th1細胞的生成來抑制主導過敏氣喘的Th2細胞,以達到治療氣喘疾病的目的,但由於Notch訊息在細胞作用上相當廣泛,如要應用到人體實驗上還需要更深入的探討。
