Abstract
近年來台灣國人十大死因中,與肥胖直接或間接相關的疾病共計有八項,而流行病學研究發現,較短的睡眠時間與身體質量指數的上升具相關性,這可能與主掌睡眠調控的褪黑激素有關。本研究旨在探討對飲食誘發血脂異常的倉鼠介入褪黑激素後是否能改善脂質代謝異常,並確認其調控脂質代謝酵素活性與相關基因表現之能力。實驗將倉鼠分為控制組與經過4 週高脂飲食誘發血脂異常之高脂組、褪黑激素低劑量(10mg/kg)、中劑量(20mg/kg)組、高劑量(50mg/kg)組,每日以管灌方式給予褪黑激素持續8週。結果顯示給予低劑量褪黑激素能降低高脂飲食所誘導之肝臟重量增加,並改善血中總膽固醇、低密度脂蛋白膽固醇濃度,同時提升高密度脂蛋白膽固醇濃度。而研究也發現介入低劑量褪黑激素能改善肝臟三酸甘油酯濃度,並使肝臟脂質合成酵素acetyl-CoA carboxylase、fatty acid synthase活性降低,而給予中劑量褪黑激素則能使脂質氧化調控相關基因carnitine palmitoyltransferase I之mRNA表現量提升。從本研究發現在飲食誘發倉鼠血脂異常的模式下可以發現褪黑激素具有降低血脂濃度、抑制肝臟脂質堆積,並調節肝臟脂質代謝路徑,進而改善高脂飲食造成的體內脂質異常代謝。
