Abstract
近年來,血小板衍生製品因其在神經保護與神經修復方面的潛在應用而受到越來越多的關注。其中,「熱處理人類血小板沉澱裂解液(heat-treated human platelet pellet lysate, HPPL)富含多種滋養因子,而經奈米過濾之 HPPL(nanofiltered HPPL, NHPPL)」則在提升病毒安全性方面具有優勢。然而,奈米過濾對 HPPL 生物學特性的影響,以及經鼻腔給藥後對成體神經新生的影響,特別是在時間動態、腦區特異性與實驗變異性等層面,仍有待進一步釐清。

本研究以小鼠模型比較經鼻腔給藥之 HPPL 與 NHPPL 對成體神經新生的影響,藉此評估奈米過濾是否可作為提升 HPPL 安全性的可行策略。分析分別於短期(第 3 天)與長期(第 28 天)進行,並透過免疫螢光染色評估與細胞增殖及細胞命運相關之標記,聚焦於齒狀迴(dentate gyrus, DG)、rostral migratory stream(RMS)及嗅球(olfactory bulb, OB)等關鍵神經新生區域。

在短期實驗中,HPPL 與 NHPPL 處理後之細胞增殖相關指標並未呈現統計學上顯著差異;然而,Ki67⁺/EdU⁺ 雙標記細胞的分布呈現輕微右移,顯示可能對過渡型增殖前驅細胞族群產生早期影響,而 EdU⁺/SOX2⁺ 細胞密度則維持相對穩定。在長期實驗中,各腦區的免疫螢光標記呈現變異性或輕度趨勢性變化,但均未達統計顯著。

上述結果可能受到多項因素影響,包括樣本數有限(每組 n = 3–4)、顯著的個體差異,以及組織取樣與定量分析上的技術限制。雖然在本研究條件下未能觀察到成體神經新生的顯著提升,但其變化方向與腦區分布整體上仍與既有文獻相符,顯示血小板衍生製品可能較偏向影響神經新生的早期階段,而非直接促進神經元成熟。

整體而言,本研究提供了 HPPL 與 NHPPL 於體內動物模型中的初步比較數據,並支持未來進一步探討奈米過濾作為推動 HPPL 朝向更安全且具臨床應用潛力之治療策略。
