Abstract
許多研究顯示,慢性飲酒會導致血液中的adiponectin蛋白質降低,進而影響肝細胞的脂質代謝,導致脂肪肝的形成與肝臟的纖維化病變。關於急性飲酒是否對肝臟與血中adiponectin濃度造成影響,則尚未有研究報導。另一方面,研究報導傳統中藥樟芝具有保護肝臟、抗氧化與抗發炎等功能;對於急性飲酒,樟芝是否具有快速有效降低血中酒精濃度等解酒的作用,對血液中adiponectin濃度又是否有影響,也是值得探討的問題。
因此本研究以大鼠為材料,實驗分成三組,樟芝(A)組(1.0 g/kg),酒精(E)組(5.5 g/kg),以及樟芝與酒精的混合液(A+E)組。分別在餵食後不同時間(0分鐘、30分鐘、60分鐘、120分鐘、240分鐘與360分鐘)進行採血,利用酵素免疫分析試劑(ELISA kits)測量血中酒精以及adiponectin濃度。結果顯示:A組血中酒精濃度在不同時間點與尚未餵食的0分鐘比較,皆沒有顯著變化。E組血中酒精濃度會逐漸上升,在120分鐘左右達到最高值(185.6 ± 31.4 mg/dL),而後逐漸下降,在360分鐘後血中酒精濃度與0分鐘的基準值比較沒有統計差異。另外,A+E組對血中酒精濃度上升的趨勢與E組並沒有明顯不同。所以結果顯示,餵食樟芝無法有效降低急性酒精餵食後上升的血中酒精濃度。
另一方面,E組在120分鐘內並不會改變血液中adiponectin的濃度;但是在240分鐘與360分鐘之後,血液中adiponectin的濃度分別約為0分鐘的0.94倍與0.95倍,有些微下降的現象,但是不具統計意義。A組對血液中adiponectin濃度沒有顯著影響。A+E組血液中adiponectin濃度在不同時間點也都沒有上升的情形。
綜合以上結果顯示,急性飲酒後,服用樟芝並不影響血液中酒精與adiponectin的濃度。因此樟芝的解酒與保肝作用,推測可能是透過保護肝臟細胞膜上的adiponectin receptor 2,避免酒精的傷害。目前正積極分析急性飲酒以及餵食樟芝後是否影響肝臟細胞的adiponectin receptor 2含量。期待能更進一步了解樟芝保護肝細胞的機轉。
