Abstract
胰臟癌由於早期無明顯症狀且缺乏可靠的診斷生物標記,至今仍是全球最致命的惡性腫瘤之一。現有的生物標記在早期檢測中的敏感性與特異性不足,凸顯出迫切需要開發新的診斷與預後標記。本研究旨在透過生物資訊分析與體外驗證模型,鑑定胰臟癌中具有臨床潛力的轉錄體生物標記。本研究利用 GEPIA2 與 UALCAN 資料庫,篩選出在胰臟腫瘤與正常組織間差異表現的基因,並鑑定出 CEACAM6、PTTG1 和 UBE2T 為潛在候選標記。CEACAM6 在腫瘤組織中表現顯著上調,並與腫瘤分級與淋巴結轉移等臨床病理參數具有高度相關性。PTTG1 與 UBE2T 則與較差的存活預後顯著相關。通路富集與網絡分析顯示,CEACAM6 參與細胞外基質(ECM)組織調控,而 PTTG1 與 UBE2T 分別與有絲分裂調控與 Fanconi 貧血通路中的 DNA 損傷修復相關。此外,UBE2C 被鑑定為在 PTTG1 與 UBE2T 網絡中共同共表現的基因,暗示其可能在腫瘤進展中扮演協同角色。透過 RT-qPCR 驗證結果顯示,CEACAM6、PTTG1 與 UBE2T 在多株胰臟癌細胞株中皆呈現上調表現,且在較具侵襲性的細胞株中,PTTG1 與 UBE2T 表現更為顯著。總結而言,這些結果指出 CEACAM6、PTTG1 與 UBE2T 為極具潛力的生物標記,未來可望應用於胰臟癌的診斷、預後評估與治療策略之發展。
