Abstract
第九型尼古丁乙醯膽鹼受體(α9-nicotinic acetylcholine receptors,簡稱α9-nAChR)會表現在人類乳房上皮細胞,並經由菸草中的成分─尼古丁,活化下游的訊息傳遞路徑並促進乳房細胞增生及癌化。根據我們過去的研究發現,乳癌檢體中α9-nAChR基因在腫瘤組織的表現量高於正常組織,且與乳癌臨床期別呈現正相關。為了瞭解乳癌轉移的分子機制,我們建立以皮下注射為基礎的原發性乳癌轉移動物模式,並取得高轉移性肺轉移乳癌細胞。經由Microarray及Real-time PCR分析發現許多過去被認為是轉移相關的基因在高轉移性乳癌細胞中顯著上升,其中也包含我們過去已有相關研究的α9-nAChR,因此本篇研究將繼續以α9-nAChR為主軸探討其對乳癌轉移的影響,並同時驗證我們所建立的原發性乳癌轉移動物模式。我們發現經由尼古丁刺激α9-nAChR,透過西方墨點法證明α9-nAChR具有促進乳癌細胞Epithelial‐to‐Mesenchymal transition的能力。利用α9-nAChR拮抗劑及專一性抗體可發現,抑制α9-nAChR可使得癌細胞在體外實驗中遷移及侵犯的能力減弱,並抑制Epithelial‐to‐Mesenchymal transition的作用。而體內實驗中,使用α9-nAChR專一性抗體可大幅降低小鼠體內乳癌轉移至肺臟的發生率。綜合本篇研究成果,我們透過一系列探討癌症轉移的實驗平台,從基因分析到細胞轉移功能分析,最後以動物模式證實了α9-nAChR與乳癌轉移的關聯性。我們也期望這些研究成果能提供未來發展α9-nAChR標靶藥物,以達到臨床上治療及預防乳癌轉移的目標。
