Abstract
本研究使用雄性自發性高血壓大鼠 (Spontaneously hypertensive rat, SHR) 以streptozotocin (STZ, 25 mg/kg BW) 誘發糖尿病,實驗動物飲食中部分蛋白質以beta-conglycinin取代,或額外添加大豆異黃酮,探討beta-conglycinin對糖尿病腎病變病程發展的影響,並釐清beta-conglycinin作用是否因蛋白質中所含大豆異黃酮影響。實驗期共進行4週,大鼠誘發糖尿病後進入實驗期,控制組 (C組) 與糖尿病腎病變組 (D組) 餵食 AIN-93M 標準飲食,beta-conglycinin 組 (B組) 飲食以AIN-93M 標準飲食為基礎,以beta-conglycinin 取代1/8 酪蛋白,大豆異黃酮組 (I組) 飲食於 AIN-93M 標準飲食額外添加0.01%大豆異黃酮(與 B 組大豆異黃酮含量相等)。實驗結果發現介入beta-conglycinin可顯著降低大鼠禁食血糖、血漿與腎臟advanced glycation end products (AGE),增加胰島素敏感性;血壓調節方面發現可有效延緩大鼠收縮壓升高,抑制angiotensin converting enzyme (ACE)活性且降低血漿與腎臟angiotensin II (Ag II)濃度,而額外添加大豆異黃酮對上述各數值皆無影響。另一方面,beta-conglycinin顯著改善尿蛋白與腎臟nephrin表現(p < 0.05),但大豆異黃酮皆無影響。結果顯示beta-conglycinin可有效延緩大鼠糖尿病腎病變病程發展,而此作用可能非來自於beta-conglycinin中所含的大豆異黃酮。
